2-Arachidonoylglycerol (2-AG) is an endocannabinoid and the most abundant endogenous ligand for cannabinoid receptors. Unlike anandamide, 2-AG acts as a full agonist at both CB1 and CB2 receptors and is produced in larger quantities. It plays roles in synaptic signaling, inflammation regulation, immune modulation, and pain pathways. 2-AG is synthesized from membrane phospholipids and degraded primarily by the enzyme MAGL (monoacylglycerol lipase). Research into 2-AG metabolism has identified MAGL inhibition as a potential therapeutic strategy for inflammation, neurodegenerative diseases, and pain management.
