Cannabis tolerance refers to the progressive reduction in response to a given dose of cannabis with repeated use, requiring higher doses for the same effect.
Tolerance to cannabis develops through downregulation and desensitization of cannabinoid receptors, primarily CB1 receptors, in response to prolonged or repeated exposure to THC. As CB1 receptors are internalized and their signaling efficiency reduces, higher doses are required to achieve the same effect. Tolerance can develop rapidly — in some studies within days of heavy use — but is reversible with abstinence, typically over 2–4 weeks. Tolerance has significant implications for medical dosing, treatment planning, and patient counseling. Cross-tolerance between cannabis and other CB1 agonists may also occur. Tolerance build-up is an important consideration in long-term medical cannabis use.
